        1. 干貨!AOM/DSS結(jié)直腸癌模型

          AOM/DSS模型是目前使用最廣泛的UC化學(xué)誘導(dǎo)模型,臨床癥狀學(xué)、形態(tài)學(xué)及病理學(xué)觀察證明,該模型與人類潰瘍性結(jié)腸炎癌變過(guò)程極為相似。AOM60751ES引起鳥(niǎo)嘌呤O6甲基化是誘導(dǎo)基因突變損傷最主要的原因,基因突變使細(xì)胞過(guò)度增殖,形成特異性的大腸腫瘤。DSS的致炎機(jī)制尚未闡明,可能與DSS的負(fù)電荷影響 DNA 合成、抑制上皮細(xì)胞增生、破壞腸黏膜屏障、導(dǎo)致巨噬細(xì)胞功能障礙及腸道菌群失調(diào)有關(guān)。

          作用機(jī)制

          偶氮甲烷(AOM)能誘導(dǎo)DNA產(chǎn)生O6-甲基鳥(niǎo)嘌加合物,導(dǎo)致G→A轉(zhuǎn)化。

           BALB/c小鼠(7周齡,雄性)

           

           

          實(shí)驗(yàn)材料

          DSS(Yeasen#60316ES,MW36000-50000)、AOM

           

           

          實(shí)驗(yàn)前準(zhǔn)備

          DSS配制:使用無(wú)菌水配制濃度1-3% DSS溶液,儲(chǔ)存在4℃?zhèn)溆谩?/span>

          AOM配制:用無(wú)菌水溶解AOM配制10 mg/mL的儲(chǔ)存液,分裝后放到-20℃保存,避免反復(fù)凍融。使用之前,融化分裝母液并用無(wú)菌生理鹽水1:10倍稀釋。

           

           

          實(shí)驗(yàn)步驟

          1.第1天稱重并標(biāo)記小鼠,實(shí)驗(yàn)組小鼠腹腔注射偶氮甲烷 AOM(10 mg/kg);
          2.用常規(guī)飲用水喂食一周;
          3.用2.5%濃度DSS替換飲用水喂食一周;
          4.用常規(guī)飲用水喂食2周;
          5.步驟3和步驟4重復(fù)3次。
