          DSS誘導果蠅潰瘍性結腸炎模型的建立

          葡聚糖硫酸鈉(Dextran Sulfate Sodium,DSS)是葡聚糖的聚陰離子衍生物,喂食果蠅會破壞腸上皮中腸道表皮細胞之間的連接從而造成腸道損傷,引起多條通路應答調控干細胞的自我更新及分化,補充受損細胞,維持腸道穩(wěn)態(tài)。

          實驗動物

          5-10日齡果蠅,雌性 

           

          實驗材料

          DSS(yeasen DSS 60316ES,MW36000-50000) 

           

          實驗步驟

          1. 用5%的蔗糖溶液配制喂食培養(yǎng)基;
          2. 對照組:5%蔗糖培養(yǎng)基(SC),實驗組:3% DSS(培養(yǎng)基配制);
          3. 預先用500 μL的喂食培養(yǎng)基將2.5cm×3.75cm的層析紙潤濕,然后放入空的培養(yǎng)瓶中;
          4. 果蠅放入含有層析紙的瓶中,29℃培養(yǎng)三天,期間每天將存活的果蠅移入含有新鮮培養(yǎng)基的空瓶中;
          5. 培養(yǎng)三天后,切開中腸,用GFP(綠色)和PH3(紅色)抗體和DRAQ5(藍色)進行免疫染色。

           通過免疫染色中不同的標記方法,量化了腸道干細胞(ISC),ISC前體細胞發(fā)生增值,表明DSS誘導果蠅模型腸道發(fā)生損傷。
